Genomic Instability in Peripheral Blood Lymphocytes of Cancer Prone Syndromes

چکیده
It is well known that several chromosomal syndromes as well as chromosomal instability syndromes are cancer prone. A thorough cytogenetic study (in certain cancer prone syndrome) that may bring out any associated factors if any in the transformation of precancerous status to cancerous status making use of both chromosomal aberrations (CA) analysis and cytokinesis block micronucleus cytome assay (CBMN cyt) was undertaken. Blood samples were collected from the cancer prone chromosomal syndromesDown syndrome (n=20); Gonadal Dysgenesis (n=5); Chromosomal instability syndromes Aplastic Anemia (n=5). Age and sex matched ethnic controls (n=30). A significantly higher frequencies of CA, micronuclei (MNi) and various nuclear anomalies were observed in the patients in comparison to the controls (p<0.001). The Aplastic Anemia patients showed a higher frequency of genomic instability followed by Gonadal Dysgenesis and Down syndrome. Therefore these biomarkers could be used to predict the risk of malignant transformation of cancer prone chromosomal syndromes.

G2 chromosomal radiosensitivity and background frequency of sister chromatid exchanges of peripheral blood lymphocytes of breast cancer patients
Background: Chromosomal alterations play an important role in carcinogenesis. Enhanced chromosomal radiosensitivity is shown for many cancer predisposition conditions including breast cancer. In this study chromosomal radiosensitivity and the frequency of background sister chromnatid exchanges (SCE) in lymphocytes of normal individuals and breast cancer patients was compared. Materials...
متن کاملComparative study of chemo-sensitivity expressed as micronuclei in lymphocytes of breast cancer patients, their unaffected first degree relatives and normal controls as a possible prognostic marker
Background: Genomic instability is one of primary causes for malignant cell transformation. In this study induced genomic instability expressed as micronuclei in breast cancer (BC) patients with different stages of the disease compared with their unaffected first degree relatives (FDR) and normal unrelated controls was investigated. Materials and Methods: The background and net micronucleus fre...
متن کاملGenomic instability and cellular stress in organ biopsies and peripheral blood lymphocytes from patients with colorectal cancer and predisposing pathologies
Inflammatory bowel disease (IBD) and polyps, are common colorectal pathologies in western society and are risk factors for development of colorectal cancer (CRC). Genomic instability is a cancer hallmark and is connected to changes in chromosomal structure, often caused by double strand break formation (DSB), and aneuploidy. Cellular stress, may contribute to genomic instability. In colorectal ...
متن کاملInterindividual differences in radiation-induced apoptosis of peripheral blood leukocytes in normal individuals and breast cancer patients
Background: Quantification of radiation-induced apoptosis in peripheral blood lymphocytes (PBLs) has been proposed as a possible screening test for cancer-prone individuals and also for the prediction of normal tissue responses after radiotherapy. Materials and Methods: The neutral version of the comet assay (single-cell gel electrophoresis) was used 24, 48, 72 hours after irradiation ...
متن کاملFoxP3+ Regulatory T Cells in Peripheral Blood of Patients with Epithelial Ovarian Cancer
Background: Ovarian cancer is the fifth leading cause of death from malignancy in women. CD4 +CD25+FoxP3+ regulatory T (Treg) cells are a subset of T lymphocytes with great inhibitory impact on immune response. Objectives: To investigate the percentage of CD4 +CD25+FoxP3+ regulatory T cells in the peripheral blood of the Iranian patients with epithelial ovarian cancer compared to healthy women ...
